To give you an overall view :)
Cardcaptor Sakura started as a manga in 1996 by a group called CLAMP, and ran until 2000, by then expanding to a 70 episode tv series, 2 movies, and tons of cds, artbooks, games, and other merchandise.General Plotline
There are two arcs: Clow Card Arc, and Sakura Card Arc. In the first one, a girl named Sakura discovers magical cards and accidentally lets them loose, and she is given the task of collecting all of them again. Then in the Sakura Card Arc, she turns all of the Clow Cards into her own cards. Thee are two movies, one after each arc, which help tie the arcs together.

The manga and anime
Just some info...
There's 12 books made by CLAMP and translated by Tokyopop into English. The first 6 books are the original story when Sakura has to capture the Clow Cards that she set free.There are only 19 cards in the manga, but the anime extends this into 53 cards (like a deck of cards, and it is sometimes used in the series sort of like tarot cards).
There are a few other differences, such as the series ending, and characters, but the anime is essentially the same as the manga.
When Cardcaptor Sakura originally came to America, Nelvana changed the name of the series to Cardcaptors, and changed the show to try to make it more compatible to both girls and boys, which didn't seem to work out too well for them. They also took out many of the love triangles.

Characters
Because they are the heart of the show :)
Kinomoto Sakura: The main character. 10 years old, a cheerful girl who grows into her role.
Kinomoto Touya: Sakura's older brother. He knows about her powers despite her efforts to hide it. He's caring and protective.
Li Syaoran: A boy who comes from China to take possession of the Clow Cards. He starts as Sakura's rival, but becomes friends fairly quickly. He is a descendant of Clow Reed.
Li Meiling: Syaoran's cousin who's also from China. She is very jealous of Sakura and challenges her to everything. She doesn't appear in the manga.
Daidouji Tomoyo: Sakura's best friend. Their mothers are cousins. She is infatuated with everything Sakura and she discovers Sakura's powers on the first day, and insists to be a part of the whole thing. She makes costumes for Sakura and Kero.

Kero-chan/Kerberos: Guardian of the Clow Book. Kero-chan fell asleep while guarding the cards, so it's also his fault the cards got away. He appointed Sakura as the cardcaptor and watches out for her. He has another form, Kerberos.

Tsukishiro Yukito/Yue: Yukito is Touya's best friend. His true form is Yue, one of Clow Reed's creations, and the one who challenges the cardcaptor when all the cards are captured. Yukito is unaware of Yue.
Hiiragizawa Eriol: Half the reincarnation of Clow Reed. He is from England and is the main antagonist in the second arc. He created Ruby Moon and Spinel Sun as rivals for Yue and Kerberos.
Clow Reed: The man who made the cards and the guardians Yue and Kerberos. He only appears a couple of times. He is reincarnated into Eriol and Sakura's father.

Suppi-chan/Spinel Sun: Like Kero-chan, he is usually in his smaller form. Although they look similar, their personalities are opposites.

Akizuki Nakuru/Ruby Moon: Yue's counterpart, created by Eriol. Nakuru has no gender, but prefers to look and act like a girl since it's cuter.
Mizuki Kaho: She is a teacher, who knows about the Clow Cards and helps Sakura from time to time. She is acquainted with both Touya and Eriol.

in reply to Sakura Madoushi shows up in the first Cardcaptor Movie (it's called Cardcaptor Sakura: The Movie. The Sealed Card is the second CCS movie). I *highly* recommend watching with the Japanese voices and reading the English subtitles to get the story CLAMP really wrote. The English dub changed the story, and IMHO, took out a lot of the emotion. If you want to see what I mean, listen to the English voices and leave the English subtitles on.
