Duet Acting Conflict & Want
There are several important things to consider when choosing an acting duet. The scene must have a conflict between the two characters in order to make the scene interesting to the audience. Each character must want something in order to propel the scene. If a character doesn't have a strong want, then why would that character engage the other? That character could choose to withdraw or leave, focusing on some other important tasks of that characters life. A strong want makes the character stay in the scene and actively engage the other participant.
The want is mostly the choice of the actor. A pair of actors can decide their own conflict and wants regardless of the dialogue in the acting duet. But, it is best to look for conflicts and wants the writer intended, since the dialogue will fit those choices. When you select a scene, look for strong conflicts and wants that occur to you as you read it.

Duet Acting Relationship
You must define the relationship of the characters. Think of how you speak to your close friends, and contrast that with how you would speak to your mother or father. There is a distinct difference, right? Now imagine how you would speak to an authority figure, like a boss, police officer, professor or teacher. In each instance you adjust your language and tone depending on who you are speaking to. The same would hold true for your character. Is the relationship of the characters husband and wife, parent and sibling, boss and employee, etc.?
Duet Acting Transititions
Another hallmark of a good acting duets are transitions. Transitions are when a character goes from one emotion or thought process to another. For example, have you ever confronted someone about something they said they would do but then didn't? Then through the course of the discussion you discovered they had a personal tragedy that preventing them from fulfilling their promise? You may have been angry or felt betrayed at the start, but you end up feeling sorry for them. In this example, you transition from anger to pity. Transitions help keep the audience engaged and add colors to a performance.
