For a guy, it is important that you don't make the move to kiss her before it's time. Moving too quickly can scare her into thinking you are only after "one thing" while taking too long can worry her that you don't like her enough to share a kiss.
Girls (and ladies) give off several different signals to indicate that she's ready to be kissed. The most obvious would be eye contact. If she is gazing into your eyes, and then glancing down at your lips, she is probably wishing you would make the move and kiss her.
